ABSTRACT

Objective To evaluate the correlation between regional blood perfusion,metabolism and angiogenesis in breast cancer.Methods The PET/CT functional imaging technique was applied to quantitatively detect the central and marginal blood perfusion parameters including blood flow(BF),blood volume(BV)and permeability of surface(Ps),and metabolism index of standard uptake value(SUV)of the tumor in 33 breast cancer patients.The expression of CD31.CD105 and VEGF in paraffin section of breast cancer were detected by immunohistochemical method,then MVD(CD31)and MVD(CD105)were obtained.The relationship between the regional blood perfusion and metabolism and MVD(CD3 1),MVD(CD105)and the expression of VEGF were analyzed. Results There was significant correlation of MVD (CD31)with BF of marginal region(P<0.05).There were significant correlations of MVD(CD105)with BF.PS and SUV(P<0.05). Conclusions Regional blood perfusion,metabolism is correlated with angiogenesis in breast cancer tissue.PETT/CT regional blood perfusion and metabolic imaging iS a noninvasive method which can be used to estimate angiogenesis status clinically in breast cancer.

ABSTRACT

Malignant tumors still threatens the human's health seriously. Metastasis and recurrence lead to the failure of treatment, and multidrug resistance adds the difficulty of therapy of to malignant tumors. But the mechanism is yet unclear. Nowadays, it is found that there's a small group of cells with selfrenewal and indefinite potency in the tumors, and they are the sources of metastasis and recurrence. This small group of cells is called cancer stem cells. Subsequently, the cancer stem ceils were separated and determined from hematological system tumor and solid tumor respectively. The cognitions for metastasis and recurrence and muhidrug resistance of malignant tumor have changed with proposal and verification of the cancer stem cell theory, which provides a new clue to the treatment of tumor and prevention for the metastasis and recurrence.

ABSTRACT

Objective To evaluate the expression of WWOX gene in breast cancer and its relation to hypermethylation of WWOX gene CpG island. Methods The expression of WWOX protein was evaluated by immunohistochemical staining in breast cancer cell line and breast cancer tissues.Methylation specific PCR(MSP)was used to check whether it Was methylated in the promoter and exon 1 of WWOX.Results Deletion in the WWOX expression was observed more frequently in invasive breast tumors,(32.2%)than normal breasttissues(5.4%)(P<0.01).20.3% of premenopause eases were completely negative for WWOX expression compared to 57.2% of postmenopause breast carcinomas(P<0.01).Furthermore,23.1% of Stage Ⅰ(6/26),28.6%of Stage Ⅱ(10/35),46.2%of Stage Ⅲ(12/26)tunlors showed negative WWOX protein expression.In breast cancer specimens.methylation rate of promoter and exon 1 CpG island of WWOX gene was 55%and 45%respectively.Whereas.WWOX CpG islands of normal mammary tissues were completely unmethylated in all cases.CpG islands of WWOX promoter and exon 1 were methylated in MDA-MB-231 cell line but not MCF-7 cell.Conclusions Some deletion in WWOX expression is common in breast cancer and methylation of WWOX DNA CpG islands plays a crucial role in the silence of WWOX.WWOX may play a role in the carcinogenesis and development of breast cancer.

ABSTRACT

Objective:To explore the effects of lipoxygenase inhibitor NDGA on telomerase and bcl-2 of breast cancer MCF-7 cells.Methods:MCF-7 cells in the logarithmic growth state were selected and measured.Four cell groups (concentration:0,1,10,100?mol/L)were divided in the experiment.After 48h treatment of NDGA,the mRNA expression of telomerase was examined by RT-PCR and the expression of bcl-2 was measured by flow cytometry.Results:After 48h treatment of different concentration of NDGA,the expressions of hTERTmRNA and bcl-2 in NDGA groups were both significantly lower than the control group (P0.001).Conclusion:NDGA can prevent proliferation and induce apoptosis in MCF-7 cells by means of the down-regulated expressions of hTERTmRNA and bcl-2.

ABSTRACT

<p><b>OBJECTIVES</b>To observe the efficacy of high intensity focused ultrasound (HIFU) in the treatment of late-stage pancreatic carcinoma and evaluate its influence on cell-mediated immunity in the host.</p><p><b>METHODS</b>Fifteen patients with late-stage pancreatic carcinoma had their tumor tissue completely destroyed with HIFU. Evaluation of efficacy was made on the basis of clinical symptom changes, variations in tumor echo, changes in pancreatic amylase, serum CA19-9 and CA242, CD3(+), CD4(+) subsets, CD4(+)/CD8(+) ratios and NK cell activity.</p><p><b>RESULTS</b>Clinical symptoms such as pain were significantly alleviated, echo of tumor was enhanced with B-US, CA19-9 and CA242 were decreased and pancreatic amylase showed no change. Eating, sleeping and mental status were all markedly improved; no serious complications were seen. On the other hand, NK cell activity was significantly enhanced in 10 patients (P < 0.05), and CD3(+) and CD4(+) subsets as well as CD4(+)/CD8(+) ratios increased to different degrees.</p><p><b>CONCLUSIONS</b>The use of HIFU in the treatment of late-stage pancreatic carcinoma is feasible and safe. It is effective in destroying the carcinoma and alleviating abdominal pain; it may enhance cell-mediated immunity in the host. This technique may offer a noninvasive therapy for the treatment of late-stage pancreatic carcinoma.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate a new operation of breast reduction for gynecomastia.</p><p><b>METHODS</b>In the past 5 years we treated 42 cases (80 sites) of gynecomastia with the double-ring incision, a supra-lateral derma-mammary pedicle of the nipple and areola and the tumescent technique.</p><p><b>RESULTS</b>The tumescent anesthesia was effective. The operation was easily performed with slight injury and less bleeding. It was safe and the patients recovered quickly. 100 to 500 grams tissue was resected from one breast. There have been no severe postoperative complications, such as nipple or areola necrosis. Follow-up from 3 to 48 months showed satisfactory results except that there was mild scar hypertrophy on the incision around the areola.</p><p><b>CONCLUSIONS</b>The new operation for gynecomastia with the double-ring incision, a supra-lateral derma-mammary pedicle of the nipple and areola, and the tumescent anesthesia is a good, reliable and convenient technique.</p>

ABSTRACT

Objective To explore the role of Bcl-2 and Caspase-3 in modulating apoptosis of ER-negative breast cancer cells induced by tamoxifen. Methods ER-negative breast cancer cell lines MDA-MB-231 were treated with 10.0?M tamoxifen for 12, 24, 36,48, 60 hours. The rate of cell apoptosis with or without caspase-3 inhibitor Ac-DEVD-CHO, and protein expression of Bcl-2,Bax were determined by flow cytometry, and the activity of Caspase-3 was examined with fluorophotometry. Results The expression of Bcl-2 was down-regulated, the activity of Caspase-3 and the rate of cell apoptosis were increased by TAM time-dependently, and the rate of apoptosis reached its peak at 48 hours. The expression of Bcl-2 was (negatively) correlated with activity of caspase-3. Tamoxifen, however, did not affect Bax protein expression. Ac-DEVD-CHO, a caspase-3 inhibitor, blocked the activation of caspase-3 and inhibited cell apoptosis (induced) by tamoxifen. Conclusions TAM could induce apoptosis in ER-negative breast cancer cells via (mitochondria) pathway by down-regulating Bcl-2 expression, and the activation of Caspase-3 might play an (important) role in the process of tamoxifen-induced apoptosis of ER-negative breast cancer cells.

ABSTRACT

Objective To study the anatomy of pelvic autonomic nerves, to avoid autonomic nerves damage during rectal cancer surgery. MethodsAnatomical dissections were carried out on 7 adult cadavers, and clinically autonomic nerve-preserving rectal cancer resection was performed. Results The superior hypogastric plexus lies just posterior to the inferior mesenteric vessels. There were no obvious autonomic nerve trunks in the loose areolar tissue plane between the parietal fascia and the visceral fascia right posterior to the rectum. There were no obvious autonomic nerve trunks between the rectum and the seminal vesicles or the prostate. The inferior hypogastric plexus was a rhomboid shaped plaque of nervous tissue. The main components of the lateral ligament were autonomic nerves passing from the pelvic plexus to the rectum within a condensation of connective tissue. WT5”HZConclusionsThe inferior mesenteric vessels can be used a landmark intraoperatively to identify the superior hypogastric plexus. In order to preserve the inferior hypogastric plexus while dissecting the lateral of rectum, we should dissect along the medial surface of the inferior hypogastric plexus, and along its curvature. ABSTRACT

Objective To evaluate the use of PET/CT in the diagnosis of breast cancer. Methods In this study,33 patients with suspicious breast tumor underwent PET/CT imaging. The images of the breast were analyzed for qualitative assessment of increased tracer uptake and blood perfusion with PET/CT. Results Among 27 cases with pathology proved breast cancer,25 was judged as PET/CT positive,2 was false-negative. Sensitivity, specificity and accuracy of PET/CT in identifying breast cancer were 92.6%,100%,93.9%. Conclusion PET/CT is a reliable and sensitive measure in the diagnosis of breast cancer in vivo.

ABSTRACT

Objective:To investigate the value of the clinical application of telom-erase activity by fine needle aspiration from breast masses for preoperative diagnosis of breast cancer in order to provide scientific basis for its practical clinical application. Methods: 85 patients with breast masses were aspirated by fine needles in the study. The samples obtained by aspiration were detected with telomerase activity by telomeric repeat amplification protocol (TRAP) assay and observed with cytomorphology by microscope respectively. The sensitivity, specificity and Youden's index of the two detective methods in diagnosis for breast cancer were compared respectively while postoperative paraffin slice pathologic results were considered as the final standard. ABSTRACT

objective At present, most operable breast cancers are confirmed by excision biopsy during the operation. If the breast cancer was definitely diagnosed by Fine -Needle Aspiration cytology(FNA) preoperatively, the excision biopsy could be avoid. Methods We diagnosed 118 inpatients of breast mass with physical examination and FNA from September, 1995 to May, 1996. Results The sensitivity and positive predict value of FNA for diagnosing breast cancer are 95.7% and 97.1%. When the palpation and FNA are both positive, the doublepositive predict value for breast cancer is 100% in our study. It is the indication of using FNA that a breast mass was diagnosed as cancer by physical examination. We can treat a breast mass with redical operation directly without excision biopsy, if the breast mass was confirmed as breast cancer by both physical examination and FNA. Conclusion We concluded that the preoperative FNA could partially replace the excision biopsy for confirming breast cancer.

ABSTRACT

Objective To study the arterial blood supply of nipple-areola and provide the anatomical basis for avoiding nipple-areola necrosis in breast operation. Methods The vascular structure of nipple-areola of 26 female breasts in 13 cadavers were studied. Results The nipple-areola mainly accepted arterial blood supply from branches of the lateral thoracic artery and the internal thoracic artery. The 2nd~4th intercostal ~perforating branches of the internal thoracic artery and branches of the lateral thoracic artery reach the base of nipple-areola from a superior,medial and upper lateral direction by passing between lobules of mammary gland, then ascend between the lacteal ducts to supply the nipple-areola; the perforators of the lateral thoracic artery and the superticial breast perforators of internal thoracic artery, formed extensive anastomoses ~subcutaneously , and particulatly under areola formed arterial rete, from which branches were given out to ~nipple-areola . The intercostal perforators and thoracoacromial perforators did not supply the nipple-areola. Conclusions When nipple-sparing mastectomy is performed, in order to avoid nipple-areola necrosis,it is necessary to protect the arterial rete under the areola, and thus, the thickness of areolar skin flap should not be less than 0.5cm; to ensure the blood supply of nipple-areola from the internal thoracic artery and the ~lateral thoracic artery in breast reduction, the superior-medial or superior-lateral breast pedicle should be used and the thickness of preserved posterior breast should not be less than 1.5cm.

ABSTRACT

Objective To observe and assess the clinicopathological significance of smooth muscle differentiation in breast cancer stromal fibroblasts. Methods A monoclonal antibody recognizing ? smooth muscle actin was used to determine the smooth muscle differentiation of fibroblasts in 69 breast cancer(BC) tissues, compared with 8 breast tissues which were removed more than 5cm from the cancer margin comfirmed without cancer cells. The relationship between the smooth muscle differentiation and clinicopathological parameters including age, tumor size, lymph node metastatic status, histological grade and ER receptor status in invasive BC were analyzed. Results Stromal fibroblasts smooth muscle differentiation was defined in 55.0% of the invasive breast cancer tissues,whereas no immunostain was demonstrated in histological normal samples. Twenty eight of the 41 cases(68.3%) with matastatic lymph nodes showed stromal fibroblasts smooth muscle differentiation,which more than that of without lymphatic metastasis group (26.3%,P

ABSTRACT

Objective: To investigate the enhancing effect of ?-interferon on anticancer effect of tamoxifen against breast cancer cells in vitro. Methods: ER-positive MCF-7 and ER-negative MDA-MB-231 breast cancer cell lines were treated with tamoxifen alone, or in combination with ?-interferon or estrogen in vitro. Cell proliferation was evaluated by MTT assay; distribution of cell cycle was determined by flow cytometry( FCM) and cell apoptosis was determined by DNA gel electrophoresis and FCM. Results: Tamoxifen inhibited the growth of ER-negtive and-positive brest cancer cells, induced G0/G1 phase arrest and induced cell apoptosis. Tamoxifen at same concentration had stronger inhibitory effect on ER-positive cells than ER-negative ones. Promoting effect of estrogen on MCF-7 growth was blocked by TAM, but inhibition of MDA-MB-231 by TAM was not related to estrogen. Anticancer effect of TAM was enhanced when cells were pre-treated with ?-interferon for 24 h. Conclusion: TAM exerts its anti-cancer effect through inhibiting proliferation and inducing apoptosis of ER-positive/negative breast cancer cells in vitro, and ?-interferon can enhance this manticancer effect of TAM on breast cancer cells.

Objective To study the expression of BRMS1mRNA in human breast cancer tissues and their significance.Methods The expression of BRMS1mRNA was detected by reverse transcription-polymerase chain reaction(RT-PCR) in 71 breast cancer tissues and adjacent breast tissues,12 patients with benign breast tumors and 12 patients with normal breast tissue,and semi-quantitative analysis of band densities was also performed.Results The expression of BRMS1mRNA in 71 patients with breast cancer and adjacent breast tissue was 0.378?0.046 and 0.918?0.044,respectively;the expression of BRMS1mRNA in 12 patients with benign breast tumors and 12 patients with normal breast tissue was 0.908?0.047 and 0.934?0.028 respectively.BRMS1mRNA expression was significantly lower in breast cancer tissue compared to adjacent breast tissue,benign breast tumors and normal breast tissue(P0.05),but was related to axillary lymph node metastasis and clinical stage(P

ABSTRACT

Objective To study the ER status in Adriamycin-sensitive and Adriamycin-resistant MCF-7 human breast cancer cell lines. MethodsThe status of ER of MCF-7/ADR and parental MCF-7 cells was detected by Western blot. The expression of ER mRNA was detected by RT-PCR .The growth and the sensitivity to Estrogen(E 2) and droloxifene(Dro) of cells were investigated by MTT assay, and the distribution of cell cycle was detected by flow cytometric assay. Results ER and ER mRNA were positive in MCF-7 cells, and negative in MCF-7/ADR cells. In comparison with MCF-7 cells, MCF-7/ADR cells showed lower growth rate, and the cell cycle was arrested at G 0/G 1 phase. E 2 at concentrations between 1?10 -12mol/L to 1?10 -7mol/L significantly stimulated the growth of MCF-7, but did not stimulate the growth of MCF-7/ADR.Dro at concentrations between 10*!?mol/L to 20*!?mol/L significantly inhabited the growth of MCF-7, and the inhibition was dose-dependent. Dro at concentrations below 20*!?mol/L did not inhibit the growth of MCF-7/ADR, dro inhabited the growth of MCF-7/ADR only at the concentration of 20*!?mol/L, and the inhibition was more effective than MCF-7. Conclusions ER was lost in MCF-7/ADR cells,probably at mRNA level. Compared with MCF-7, the growth rate of MCF-7/ADR decreased,MCF-7/ADR cells lost the dependence on estrogen and the sensitivity to endocrine therapy.
